The official Autobiography of Mark Twain (first published in 1924) is available at public libraries but remains in copyright limbo for reasons explained in the Wikipedia article Autobiography of Mark Twain.  

The official (1924 copyright) autobiography has been recently revised updated and annotated based on dictaphone recordings left behind by Twain. This Mark Twain Project version is now available for online reading at the following links:

The Autobiography of Mark Twain Summary, a useful introduction and overview of the 1924 autobiography (which is certainly available in academic libraries) may be found at eNotes.com. 

Several chapters (earlier versions) of Twain's autobiography were published independently before 1910 in the pages of the North American Review, and these magazine editions are considered to be public domain.
